Integrin에 의한 TGF-b signaling 저해제 개발 업체다. 이쪽을 건드리다보니 적응증은 대부분 fibrosis 계열.

PLN-74809 (small molecule)

Lead Asset인 PLN-74809는 avb6 / avb1 intergin dual selective inhibitor로 IPF 적응증에서 개발 중이다. 1a SAD/MAD study, 1b POC는 완료하였고 현재 2개의 2a trial의 enrollment를 진행 중이다. IPF는 Esbriet (pirfenidone, Genetech)과 Ofev(nintedanib, BI) 이외에는 약이 없는 상황.

'20년 3월에는 PSC (primary sclerosing cholangitis, 경화성 담관염) 적응증의 IND filed, 하반기 중 개시 예정. 

PLN-1474 (small molecule)

intergin avb1 selective inhibitor로 NASH-associated liver fibrosis 적응증으로 1상 SAD/MAD 진행 중. '20년말 top line data 예정되어 있으며, NVS와 이미 파트너십을 맺고 비용 다 대주고 있고, 끝나면 권리를 가져갈 수 있게 되어있다.

그 외에도 integrin-based 초기 약물들이 몇개 있다. 

비슷한 컨셉으로 oral, selective integrin inhibitor를 개발하고 있는 MORF가 있다. 여기는 a4b7(IBD)가 메인이고, PLRX와 동일하게 avb6(IPF/PSC)를 ABBV와 개발 중이다. dual이냐(PLRX) single이냐(MORF)의 차이인데, 같이 보면 재미있을 듯.
